{"id":40570,"date":"2024-11-03T21:33:08","date_gmt":"2024-11-03T20:33:08","guid":{"rendered":"https:\/\/otpor.media\/?p=40570"},"modified":"2024-11-03T21:33:17","modified_gmt":"2024-11-03T20:33:17","slug":"novo-otkrice-procesa-popravka-dna","status":"publish","type":"post","link":"https:\/\/otpor.media\/?p=40570","title":{"rendered":"Novo otkri\u0107e procesa popravka DNA"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\">Me\u0111unarodni tim istra\u017eiva\u010da sa Sveu\u010dili\u0161ta Oxford (Oxford), Tehnolo\u0161kog sveu\u010dili\u0161ta Nanyang u Singapuru (NTU Singapore) i Instituta Ru\u0111er Bo\u0161kovi\u0107 (IRB) otkrio je novi proces popravka o\u0161te\u0107ene DNA koji bi mogao zna\u010dajno utjecati na lije\u010denje raka i zdravlje stanica.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Rezultati istra\u017eivanja objavljeni su u \u010dasopisu Cell (Lescaux et al., 2024).<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">UV zra\u010denje, kemikalije iz okoli\u0161a, pa \u010dak i prirodni procesi unutar stanice mogu uzrokovati o\u0161te\u0107enje DNA, molekule koja nosi sve informacije potrebne za funkcioniranje \u017eivota. Stoga, stanice u na\u0161em tijelu svakodnevno prolaze kroz niz &#8220;popravaka&#8221; kako bi osigurale pravilno funkcioniranje. No, \u0161to se doga\u0111a kada ti popravci zaka\u017eu?<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">&#8220;Putevi popravka o\u0161te\u0107enja DNA klju\u010dni su mehanizmi koje stanice koriste za odr\u017eavanje integriteta svojih genetskih informacija. Ovi mehanizmi poma\u017eu popraviti o\u0161te\u0107enja i sprije\u010diti \u0161tetne mutacije koje mogu dovesti do pojave tumora, ili do stani\u010dne smrti. Stani\u010dna smrt je s jedne strane po\u017eeljan ishod, jer stanica optere\u0107ena mutacijama mo\u017ee izazvati te\u0161ke bolesti poput raka, dok je s druge strane nepo\u017eeljan ishod, ako se radi o neuronima jer mo\u017ee dovesti do bolesti mozga poput demencije ili neurodegenerativnih bolesti&#8221;, obja\u0161njava dr. sc. Marta Popovi\u0107, voditeljica Grupe za DNA popravak u Laboratoriju za molekularnu ekotoksikologiju Zavoda za istra\u017eivanje mora i okoli\u0161a IRB-a.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Jo\u0161 od 1950-ih godina kada je otkrivena struktura DNA i prvi putevi popravka DNA o\u0161te\u0107enja, smatralo se da se popravak DNA uvijek doga\u0111a na mjestu o\u0161te\u0107enja, a to su stani\u010dna jezgra i mitohondriji u kojima se DNA nalazi. No, novo istra\u017eivanje objavljeno u \u010dasopisu Cell pokazuje kako se proces popravka DNA mo\u017ee odvijati izvan jezgre, u citoplazmi, uz pomo\u0107 procesa poznatog kao autofagija. To je prirodni proces recikliranja stani\u010dnih dijelova, tijekom kojeg se &#8220;stani\u010dni otpad&#8221; \u0161alje u lizosome &#8211; organele odgovorne za razgradnju i recikliranje molekula koje vi\u0161e nisu potrebne stanici.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Istra\u017eivanje koje je proveo ovaj multidisciplinarni tim znanstvenika pro\u0161iruje dosada\u0161nje spoznaje o autofagiji, uvode\u0107i novi proces nazvan nukleofagija. Dok je autofagija ve\u0107 poznata kao proces kojim stanice razgra\u0111uju vlastite o\u0161te\u0107ene dijelove, nukleofagija specifi\u010dno uklju\u010duje uklanjanje o\u0161te\u0107enih dijelova DNA iz jezgre. U ovom procesu, proteini koji su nepovratno kovalentno vezani na DNA (eng. DNA-protein crosslinks &#8211; DPC), uklanjaju se iz jezgre i transportiraju do lizosoma gdje se razgra\u0111uju. Na taj na\u010din nukleofagija \u0161titi genetski materijal stanica i doprinosi njihovom pre\u017eivljavanju.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Multidisciplinarni pristup ovog istra\u017eivanja bio je klju\u010dan za otkri\u0107e nukleofagije, a uklju\u010divao je biokemiju, stani\u010dnu biologiju, masenu spektrometriju, bioinformatiku, kori\u0161tenje ribe zebrice kao \u017eivotinjskog modela i materijale pacijenata oboljelih od raka debelog crijeva.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Centralni protein u ovom procesu je TEX264. On ima tri uloge: prepoznaje DPC o\u0161te\u0107enja i dovodi druge proteine potrebne na popravak na mjesto lezije, zatim pokre\u0107e nukleofagiju tako da omogu\u0107uje izlazak DNA o\u0161te\u0107enja kroz jezgrinu ovojnicu, i naposljetku omogu\u0107uje ulazak o\u0161te\u0107enja u lizosome tako \u0161to stupa u interakciju s LC3 receptorom u membrani lizosoma.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Ovo otkri\u0107e ima klju\u010dnu va\u017enost u kontekstu istra\u017eivanja tumora, jer neki kemoterapijski lijekovi, kao \u0161to je irinotekan, uzrokuju stvaranje DPC o\u0161te\u0107enja. TEX264 omogu\u0107uje stanicama da uklone ova o\u0161te\u0107enja i pre\u017eive terapiju, \u0161to zna\u010di da tumorske stanice mogu uspje\u0161no obnoviti svoj genetski materijal i tako nastaviti rasti.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Idu\u0107i korak je razvoj lijekova koji bi mogli blokirati TEX264 i sprije\u010diti tumorske stanice u popravljanju o\u0161te\u0107enja. Kombiniranjem inhibicije TEX264 proteina s postoje\u0107im lijekovima mogli bi dodatno oslabiti tumorske stanice i pobolj\u0161ati u\u010dinkovitost kemoterapije. Ova strategija mogla bi donijeti zna\u010dajna pobolj\u0161anja u lije\u010denju raka debelog crijeva i drugih tumora.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Kako bi istra\u017eili funkciju nukleofagije i proteina TEX264 na razini organizma, znanstvenici su koristili zebricu (Danio rerio), malenu ribu \u010diji je genom vrlo sli\u010dan ljudskom. Zebrice su idealni model za biomedicinska istra\u017eivanja jer omogu\u0107uju jednostavno pra\u0107enje stani\u010dnih procesa u stvarnom vremenu. Njihova vanjska oplodnja i razvoj embrija omogu\u0107uju lak\u0161e prou\u010davanje stani\u010dnih mehanizama, dok genetske manipulacije pomo\u0107u CRISPR-Cas sustava omogu\u0107uju istra\u017eiva\u010dima relativno brzo i precizno modificiranje gena.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Tim s Ru\u0111era, dr. sc. Cecile Otten i dr. sc. Ivan Anti\u010devi\u0107 pod voditeljstvom dr. sc. Marte Popovi\u0107 iz Grupe za DNA popravak u Laboratoriju za molekularnu ekotoksikologiju Zavoda za istra\u017eivanje mora i okoli\u0161a pokazao je da uti\u0161avanje gena TEX264 uzrokuje akumulaciju DNA o\u0161te\u0107enja u embrijima zebrice. Znanstvenici su tako\u0111er otkrili da, nakon ponovne aktivacije TEX264, DNA normalno zapo\u010dinje popravak. Nasuprot tome, ako u embrijima zebrice aktiviraju TEX264 protein koji nosi mutaciju u domeni LIR, odgovornom za izlazak o\u0161te\u0107enja iz jezgre i njihov ulazak u lizosom, popravak je onemogu\u0107en, \u0161to pokazuje da je ovaj protein klju\u010dan upravo za proces nukelofagije.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Nakon eksperimenata na zebrici, kada je postalo jasno da je popravak DNA pomo\u0107u TEX264 proteina u procesu nukleofagije bitan i na razini organizma, grupa dr. sc. Kristijana Ramadana sa Sveu\u010dili\u0161ta u Oxfordu istra\u017eila je da li procesi autofagije direktno popravljaju o\u0161te\u0107enu DNA nakon kemoterapije. Analize tumora debelog crijeva na 300-tinjak pacijenata koji su primali standardne terapije pokazale su da na osnovu koli\u010dine TEX264 proteina u stanicama raka debelog crijeva mo\u017eemo predvidjeti kako \u0107e pacijenti odgovoriti na terapiju i produ\u017eiti efikasnost standardne terapije.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Ovi rezultati otvaraju vrata razvoju novih lijekova koji ciljano djeluju na proces nukleofagije i TEX264 protein, omogu\u0107uju\u0107i u\u010dinkovitije lije\u010denje tumora. Kori\u0161tenje zebrice kao modela u istra\u017eivanjima dodatno nagla\u0161ava va\u017enost ove ribe u biomedicinskim istra\u017eivanjima zbog njezine genetske sli\u010dnosti s ljudima i jednostavnosti za laboratorijske eksperimente.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Upravo s ciljem prou\u010davanja ljudskih bolesti i toksikolo\u0161kih procesa, Laboratorij za molekularnu ekotoksikologiju pri Zavodu za istra\u017eivanje mora Instituta Ru\u0111er Bo\u0161kovi\u0107 uspostavio je pogon za zebrice. Istra\u017eiva\u010dke grupe pod vodstvom dr. sc. Tvrtka Smitala i dr. sc. Marte Popovi\u0107 razvile su postupke izmjene genoma pomo\u0107u CRISPR-Cas tehnologije te razvijaju nove sojeve zebrica za biomedicinska i toksikolo\u0161ka istra\u017eivanja.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Za razliku od mi\u0161eva i \u0161takora, zebrice pru\u017eaju brojne prednosti: vanjska oplodnja i razvoj omogu\u0107uju pra\u0107enje pod mikroskopom, dok ve\u0107i broj jaja\u0161aca po leglu osigurava statisti\u010dki zna\u010dajnije rezultate. Osim toga, njihovo odr\u017eavanje je jednostavnije i jeftinije. Zebrice dijele 99% gena uklju\u010denih u popravak DNA s ljudima, dok 84% ljudskih gena povezanih s bolestima imaju svoje ekvivalente kod zebrica, \u0161to ih \u010dini izvrsnim modelom za istra\u017eivanja raka, neurodegenerativnih, kardiovaskularnih i metaboli\u010dkih bolesti.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Tako\u0111er bitno, zamjena modela glodavaca zebricama u skladu je sa smjernicama EU komisije za kori\u0161tenje \u017eivotinjskih modela u znanstvenim istra\u017eivanjima i slijedi na\u010delo 3R: Reduce, Replace, Refine.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><a href=\"https:\/\/www.plivazdravlje.hr\/vijesti\/clanak\/38556\/Novo-otkrice-procesa-popravka-DNA.html\">PlivaZdravlje<sup>.hr<\/sup><\/a><\/strong><\/p>\n","protected":false},"excerpt":{"rendered":"<p>Me\u0111unarodni tim istra\u017eiva\u010da sa Sveu\u010dili\u0161ta Oxford (Oxford), Tehnolo\u0161kog sveu\u010dili\u0161ta Nanyang u Singapuru (NTU Singapore) i Instituta Ru\u0111er Bo\u0161kovi\u0107 (IRB) otkrio je novi proces popravka o\u0161te\u0107ene DNA koji bi mogao zna\u010dajno utjecati na lije\u010denje raka i zdravlje stanica.
